Schisandrin B exerts anticancer effects on human gastric cancer cells through ROS-mediated MAPK, STAT3, and NF-κB pathways

Schisandrin B (Sch B) is a monomer with anti-cancer and anti-inflammatory effects, which are isolated from the plant Schisandra chinensis (Turcz) Baillon. We investigated anti-gastric cancer (GC) effects of Sch its underlying molecular mechanisms. The Cell Counting Kit-8 assay was used to determine on viability GC normal cell lines. Hoechst/propidium iodide staining flow cytometry were assess apoptosis induction B. Western blotting evaluate downstream apoptotic proteins. DCFH-DA fluorescent probe regulatory reactive oxygen species (ROS) levels related signaling pathways in cells. results showed that could regulate phosphorylation level mitogen-activated protein kinase (MAPK) by upregulating ROS accumulation gastric cells, then reduce expression nuclear factor kappa (NF-κB) phosphorylated transcription 3 (p-STAT3). In addition, downregulated cycle proteins cyclin-dependent 2/4/6 cyclin D1/E, arrested cells G0/G1 phase.
